General Objective: The General objective of proposed study is to determine the effects of Berberine on molecular signaling of activated T-Cells in treatment of autoimmune Diabetes
Specific objectives:
1) To assess the Berberine on decreasing of glucose level in blood
2) To determine the effectiveness of Berberine on expression of genes IFNγ, IL-17, and RORγt using Real-Time quantitative PCR
3) To determine the effectiveness of berberine on differentiation of CD4+ T-cells
4) To determine the effectiveness of berberine on activating of proteins p38MAPK, JNK, ERK 1/2, STAT1, and STAT4 using western blotting and ELISA
Hypotheses:
1) Berberine can reduce glucose level of diabetic mice.
2) Berberine can inhibit expression of genes involved in cell signaling pathways of helper T-cells.
3) Berberine can inhibit differentiation of CD4+ T-cells.
4) Berberine can inhibit phosphorylation of proteins involved in cell signaling pathways of helper T-cells.
